Biologics (2) IL-12/23 Inhibitors 生物制剂之白细胞介素-12/23抑制剂

What is Interleukin 12 (IL-12)

Interleukin 12 has two subunits: IL-12A (p35) and IL-12B (p40). The IL-12 is produced by macrophages, monocytes and B lymphocytes to against infections such as AIDS, malaria, and tuberculosis. The macrophages, monocytes and B lymphocytes are kinds of immune system white blood cells, they locate and ‘eat’ harmful particles such as bacteria, viruses, fungi, and parasites. [1,2]

IL-12 is essential for protective immunity to intracellular bacteria such as mycobacteria and salmonella. [3]

Impaired IL-12 causes immunodeficiencies. [4]
Elevated IL-12 is linked with immune-mediated diseases, psoriasis, psoriatic arthritis, Crohn’s disease, multiple sclerosis. [5]

What is Interleukin 23 (IL-23)

Interleukin 23 has two subunits: IL-23A (p19) and IL-23B (p40). The IL-23 is produced by dendritic cells and macrophages. The macrophages and dendritic cells are kinds of immune system white blood cells, they locate and ‘eat’ harmful particles such as bacteria, viruses, fungi, and parasites.

Elevated IL-23 is linked with immune-mediated diseases, psoriasis. [6]

Ustekinumab (Stelara ® ) is a monoclonal antibody that targets IL-12 and IL-23, whereas guselkumab (Tremfya TM ), risankizumab, and tildrakizumab are monoclonal antibodies that target IL-23. [7]
